1. Results of sequencing
After transformation of the BioBrick BBa_K593004 and cultivation in E. coli the plamid DNA was isolated. We determined the nucleotide sequence of the BioBrick BBa_K593008 by use of sequencing techniques. The alignment of BBa_K593008 with the original sequence of mmoC fromMethylococcus capsulatus shows that they differed significantly (see Fig.1).

The tested BioBrick does not contain the sequence of mmoC. The DNA sequence is without any distinct function. A further characterization of the BioBrick BBa_K593008 was impossible. We were not able to perform any expression studies or activity tests with this BioBrick.
2. Improvement
Our new BioBrick BBa_K1390002 contains the original sequence of mmoC from Methylococcus capsulatus, as shown in the alignment of our new Brick and the original sequence from Methylococcus capsulatus (see Fig.2).
